Targeting Chromatin Remodeling Complexes in Cancer
The mammalian SWI/SNF, or BAF, chromatin remodeling complex has been implicated as a therapeutic target in cancer, although the most effective way to target this heterogeneous multisubunit complex is unclear. We are defining the biochemical composition of SWI/SNF complexes in various cell types and establishing the specific subunits required for relevant disease phenotypes. Then we are deciphering how various chromatin targeting domains, such as bromodomains, chromodomains and plant homeodomains, contribute to the overall function of SWI/SNF subcomplexes. From this information, we have defined new targets and new strategies for small molecule inhibitor development.
